很多老師使用瑞士insphero 的3D懸滴培養(yǎng)板獲得3D微組織后,要提取RNA進(jìn)行熒光定量PCR。下面這篇文章就是介紹如何提取insphero 3D微組織中的RNA:
使用Promega ReliaPrep RNA Tissue Miniprep System提取三維(3D)微組織的RNA
Introduction介紹
基于細(xì)胞的藥物開發(fā)和毒理學(xué)的多項(xiàng)分析,從培養(yǎng)細(xì)胞中提取高質(zhì)量的RNA是非常重要的。一些應(yīng)用如實(shí)時(shí)熒光定量PCR或微陣列分析的前提都是需RNA提取步驟。下面介紹的是從很小的微組織(約1000個(gè)細(xì)胞)中提取總RNA的protocol(ReliaPrep RNA Tissue Miniprep System, promega)。
若想使用自動(dòng)化RNA提取方法:可使用Promega的Maxwell 16 System。
Summary概述
從3D Insight 人肝臟微組織中提取RNA(ReliaPrep RNA Tissue Miniprep System)。提取的RNA樣本用QuantiFluor RNA system,Agilent 2100 Bioanalyzer,GoTaq 1-Step RT-qPCR System進(jìn)行分析。
Materials材料
3D Insight Microtissues in GravityTRAP(Insphero)
1×PBS,含鈣離子和鎂離子
ReliaPrep RNA Tissue Miniprep System(Promega Z6110)
Quantus Fluorometer(Promega E6150)
QuantiFluor RNA system(Promega E3310)
Procedure操作流程
一、微組織的準(zhǔn)備
1.輕柔的吸除GraityTRAP板中的培養(yǎng)基,用70ul的1×PBS洗滌微組織2次,如下圖Figure 1:

注:輕柔、緩慢洗滌微組織(<30ul/秒)
2.轉(zhuǎn)移微組織,1000ul吸頭垂直覆蓋GraityTRAP板孔的底端,吸頭要與孔底部緊密接觸,如Figure 2,將微組織吸出(約50ul)轉(zhuǎn)移到1.5ml離心管中,避免吸入氣泡(避免微組織損失)。

3. 4000g離心1min,去除PBS,直接作為RNA提取的樣本
二、RNA提取—ReliaPrep RNA Tissue Miniprep System
1.直接加250ul LBA+TG buffer到樣本中
2.漩渦震蕩10秒,用吸頭吹打10次,裂解微組織。如想更*裂解,可使用掌上均質(zhì)器5-10秒
3.再加85ul的isopropanol(異丙醇),漩渦震蕩5秒混合均勻
4.再參照ReliaPrep RNA Tissue Miniprep System的手冊(cè)進(jìn)行結(jié)合和洗脫等步驟
5.用30ul水洗提RNA后,冰上放置
6.RNA濃度和產(chǎn)量用QuantiFluor RNA system和Quantus Fluorometer檢測(cè)
Typical Results結(jié)果
使用ReliaPrep RNA Tissue Miniprep System提取3D InSight人肝臟微組織中的RNA,結(jié)果見Figure 3.
Figure 3:A-RNAs were isolated from either 1,2,4,or 8 microtissues. Yields were determined using the QuantiFluor RNA system with the Quantus Fluorometer and the RNA 6000 Pico kit. All values are greater than 9 indicating high quality RNA.
B-HPRT specific primers were used with the GoTaq 1-Step RT-qPCR System to show amplifiability of the isolated RNAs.
C-RNAs were isolated from single microtissues to compare the average yield from one microtissue using this purification system.The line represents the average yield(n=6).
